TÉCNICAS

ANALYTICAL TECHNIQUES

  • Liquid chromatography (analytical and preparative conditions)
  • Liquid chromatography coupled to tandem mass spectroscopy. Available equipment in the group:
    • Ion trap (IT) mass spectrometer Bruker Esquire 3000, coupled to a liquid cromatography system Agilent Series 1100
    • Quadrupole-time of flight (Q-TOF) mass spectrometer Bruker Microtof QII, coupled to a Waters Acquity UPLC
    • MALDI-TOF/TOF mass spectrometer Bruker Autoflex speed, nano chromatography system and fraction collector for MALDI plates (Proteineer fcII).
  • Gel electrophoresis (inmunoblotting, in-gel digestion and mass spectrometry analysis) and capillary electrophoresis
  • Immunoenzymatic techniques (Western-Blot, ELISA, ELLA)
  • Spectrometric and fluorimetric techniques
  • Quantitative real time PCR (gene expression in cellular and animal models)
  • Flow cytometry
  • Sensory analysis
  • Blood pressure measurement in animal models (indirect method, tail cuff)

CELLS AND TISSUES

  • Cell viability and cycle studies. Apoptosis assays
  • Biomarkers of oxidative stress, allergy, inflammation and carcinogenesis
  • Studies on immune response against food allergens
  • Vascular reactivity studies
  • Cell lines:
    • Mouse macrophages RAW264.7
    • Rat intestine IEC-6 cells
    • Hamster lung V79 cells
    • Human colon (HT-29 y Caco2), prostate (PC-3), lung (A-549), stomach (AGS), liver (HepG2), and ovary (Caov-3) cancer cells
  • Primary cell cultures:
    • Human peripheral blood mononuclear cells (PBMC) and derived subtypes (polymorphonuclear cells, monocytes, etc)
    • Spleen, mesenteric lymph nodes, Peyer´s patches and peritoneal macrophage cell cultures from Balb/C and C3H/HeOuJ mice
    • Aorta vascular smooth muscle and fibroblast cell cultures from rat

ANIMAL MODELS

  • Mouse models of oral and intraperitoneal sensitization to food allergens (egg, milk, peanut) using BALB/c and C3H/HeOuJ strains
  • Mouse models of anaphylaxis against food allergens (egg, milk, peanut) with BALB/c and C3H/HeOuJ strains
  • Rat models of obesity, type II diabetes and metabolic syndrome induced by the diet
  • Rat models of toxicity by heavy metals to study cardiometabolic complications linked to oxidative stress
  • Wistar, Sprague-Dawley (SD), spontaneously hypertensive (SHR), Wistar-Kyoto (WKY) and genetically obese (Zucker) rats, to evaluate functional foods and ingredients with beneficial effects on cardiometabolic health
